D8 Biomarkers in blood and CSF (tau, amyloid, neurofilaments)
Title
Fluid biomarkers are quantitative measurements of specific molecules in blood or CSF that directly reflect the underlying pathological processes described in series A. The concentration of beta-amyloid 42 and its ratio to amyloid 40 are derived directly from the aggregation kinetics of Abeta introduced in the Alzheimer's model (A3), as is plasma phosphorylated tau (p-tau), which reflects the variable tau_p of the same model. Neurofilament light chain (NfL) is a nonspecific but sensitive marker of axonal damage, directly extending the cytoskeletal integrity variable used in TBI (A6) and ALS (A10). Each biomarker also follows its own kinetics of release and clearance in the blood, modeled as a simple compartmental system. The diagnostic performance of a biomarker is formally evaluated by its sensitivity and specificity depending on the chosen cut-off point, summarized in the ROC curve and its area under the curve.
